The canonical, four-phase model of the cell cycle has dominated our thinking for over seventy years. This model is still useful but falls woefully short of capturing the sophisticated mechanisms that regulate cell cycle progression and arrest. Drawing on recent advances in single-cell microscopy, I will share our lab's progress in measuring cell cycle states, building computational models of cell cycle progression and arrest, and exploiting these approaches to understand drug resistance in cancer.
